Will converting to JPG lose quality?

JPEG is lossy, but at the quality used here the difference is invisible at normal viewing sizes. For photographs the size saving is enormous compared with the quality cost.

What happens to transparency?

It is flattened onto a white background, because JPEG cannot store an alpha channel. Keep the file as PNG if transparency matters.

Should I convert screenshots to JPG?

No. JPEG blurs sharp edges and text, which is exactly what a screenshot is made of. Compress those as PNG instead.
